Carcinogenesis Mutagenesis Impairment Of Fertility
Long-term animal studies have not been performed to evaluate the carcinogenic potential of the combination of clotrimazole and betamethasone dipropionate or either component individually.
Betamethasone was negative in the bacterial mutagenicity assay and in the mammalian cell mutagenicity assay . It was positive in thein vitro human lymphocyte chromosome aberration assay, and equivocal in the in vivo mouse bone marrow micronucleus assay.
Reproductive studies with betamethasone dipropionate conducted in rabbits at doses of 1.0 mg/kg/day by the intramuscular route and in mice up to 33 mg/kg/day by the intramuscular route indicated no impairment of fertility except for dose-related increases in fetal resorption rates in both species.
In a combined study of the effects of clotrimazole on fertility, embryofetal development, and postnatal development, male and female rats were dosed orally with dose levels of 5, 10, 25, or 50 mg/kg/day from 10 weeks prior to mating until 4 weeks postpartum. No adverse effects on the duration of estrous cycle, fertility, or duration of pregnancy were noted.

How Is This Drug Best Given
Give this drug as ordered by your childs doctor. Read all information given to you. Follow all instructions closely.
All skin products:
- Keep using this drug as you have been told by your childs doctor or other health care provider, even if your childs signs get better.
- Wash your hands before and after use.
- Do not give by mouth. Use on your childs skin only. Keep out of your childs mouth, nose, and eyes .
- Clean affected part before use. Make sure to dry well.
- Put a thin layer on the affected part and rub in gently.
- Do not use coverings unless told to do so by the doctor.
Vaginal cream:
- Keep using this drug as you have been told by your childs doctor or other health care provider, even if your childs signs get better.
- Use at bedtime for best results.
- Wash your hands before and after use.
- Do not give this drug by mouth. For vaginal use only. Keep out of your childs mouth, nose, and eyes .
- Dry the outside vaginal area all the way after your child showers, bathes, or swims. Be sure your child does not swim for 9 to 12 hours after using this drug. Have your child change out of wet bathing suits or damp workout clothes right away.
If your child is or may be sexually active:
- Vaginal sex must be avoided while using this drug.
About Clotrimazole For Thrush
Clotrimazole is an antifungal medicine.
It’s used to treat yeast infections including thrush in women and men, although thrush is more common in women.
Clotrimazole comes as an external cream, an internal cream and a pessary .
For vaginal thrush you can use pessaries or cream or both at the same time. The pessaries and internal cream are sometimes sold together.
Clotrimazole is available to buy in pharmacies and shops. Some stronger treatments are only available with a prescription.

Who Can And Cannot Use Clotrimazole
Clotrimazole creams can be used by most adults and children.
Clotrimazole may not be suitable for some people. To make sure clotrimazole is safe for you, tell a pharmacist or doctor if:

- you’ve had thrush more than twice in the past 6 months
- you or your partner have ever had a sexually transmitted infection

Indications And Usage For Clotrimazole Betamethasone Cream
Clotrimazole and betamethasone dipropionate cream is a combination of an azole antifungal and corticosteroid and is indicated for the topical treatment of symptomatic inflammatory tinea pedis, tinea cruris, and tinea corporis due to Epidermophyton floccosum, Trichophyton mentagrophytes, and Trichophyton rubrum in patients 17 years and older.

Effects On Endocrine System
Clotrimazole and betamethasone dipropionate cream can cause reversible hypothalamic-pituitary-adrenal axis suppression with the potential for glucocorticosteroid insufficiency. This may occur during treatment or after withdrawal of treatment. Cushings syndrome and hyperglycemia may also occur due to the systemic effect of corticosteroids while on treatment. Factors that predispose a patient to HPA axis suppression include the use of high-potency steroids, large treatment surface areas, prolonged use, use of occlusive dressing, altered skin barrier, liver failure, and young age.
Because of the potential for systemic corticosteroid effects, patients may need to be periodically evaluated for HPA axis suppression. This may be done by using the adrenocorticotropic hormone stimulation test.
In a small trial, clotrimazole and betamethasone dipropionate cream was applied using large dosages, 7 g daily for 14 days to the crural area of normal adult subjects. Three of the 8 normal subjects on whom clotrimazole and betamethasone dipropionate cream was applied exhibited low morning plasma cortisol levels during treatment. One of these subjects had an abnormal cosyntropin test. The effect on morning plasma cortisol was transient and subjects recovered 1 week after discontinuing dosing. In addition, 2 separate trials in pediatric subjects demonstrated adrenal suppression as determined by cosyntropin testing .

How To Use Clotrimazole
If you are using the over-the-counter product to self-treat, read all directions on the product package before using this medication. If you have any questions, consult your pharmacist. If your doctor has prescribed this medication, use it as directed.
Read the Patient Information Leaflet available with this product. Consult your doctor or pharmacist if you have any questions.
This product is for vaginal use only. Wash your hands before and after use. Avoid contact of this product with your eyes. If it gets into your eyes, wash them right away with plenty of water. Call your doctor if eye irritation lasts.
Dosage is based on your medical condition and response to treatment. Use the medication usually once daily at bedtime for 1 to 7 nights depending on the product used, or as directed by your doctor.
If using the vaginal cream, follow the package instructions on how to fill/use the applicator with the cream. Lie on your back with your knees toward your chest. Insert one applicator filled with cream into the as far as it will comfortably go. Slowly press the plunger of the applicator in to apply the cream. Make sure to insert the correct cream for use inside the , not the skin cream for use on the outer genital area .
Continue to use this medication every day for the full time prescribed, even if symptoms disappear after a few days or if your menstrual period starts. Stopping the medication too soon may allow the yeast infection to return.

What Form Does This Medication Come In
Each 1 g of smooth white cream contains 10 mg of clotrimazole USP and 0.64 mg of betamethasone dipropionate, USP, equivalent to 0.5 mg of betamethasone USP, in a hydrophilic emollient cream. Nonmedicinal ingredients: purified water, cetostearyl alcohol, white petrolatum, mineral oil, cetomacrogol 1000, phosphoric acid, monobasic sodium phosphate, propylene glycol, benzyl alcohol as preservative and sodium hydroxide to adjust the pH.
How Should I Use This Medicine
This cream is for external use only. Do not take by mouth. Follow the directions on the prescription label. Wash your hands before and after use. If treating hand or nail infections, wash hands before use only. Apply a thin layer of cream to the affected area and rub in gently. Do not cover or wrap the treated area with an airtight bandage . Use the cream for the full course of treatment prescribed, even if you think the condition is getting better. Use the medicine at regular intervals. Do not use more often than directed. Do not use on healthy skin or over large areas of skin. Do not use this medicine for any condition other than the one for which it was prescribed. When applying to the groin area, apply a small amount and do not use for longer than 2 weeks unless directed to by your doctor or health care professional. Do not get this cream in your eyes. If you do, rinse out with plenty of cool tap water.
Talk to your pediatrician regarding the use of this medicine in children. While this drug may be prescribed for children as young as 17 years for selected conditions, precautions do apply.
Patients over 65 years old may have a stronger reaction and need a smaller dose.
Overdosage: If you think you have taken too much of this medicine contact a poison control center or emergency room at once.

What Should I Watch For While Using This Medicine

It may also decrease the effect of vaginal spermicides. Do not rely on any of these methods to prevent sexually transmitted diseases or pregnancy while you are using this medicine.

Clotrimazole And Betamethasone Dipropionate
Clotrimazole and Betamethasone Dipropionate Cream, USP has been shown to be at least as effective as clotrimazole alone in a different cream vehicle. No comparative studies have been conducted with Clotrimazole and Betamethasone Dipropionate Lotion and clotrimazole alone. Use of corticosteroids in the treatment of a fungal infection may lead to suppression of host inflammation leading to worsening or decreased cure rate.
How Should I Use This Medication
Apply a thin film of cream to completely cover the affected and surrounding skin areas twice daily, in the morning and at night or as directed by your doctor. The cream is usually used for a period of 2 to 4 weeks depending on the condition being treated. Using the cream for more than 4 weeks is not recommended.
This cream should not be covered with bandages or other coverings after application. Do not use this medication in or near the eyes.
Many things can affect the dose of medication that a person needs, such as body weight, other medical conditions, and other medications. If your doctor has recommended a dose different from the one above, do not change the way that you are using the medication without consulting your doctor.
It is important to use this medication exactly as prescribed by your doctor. If you miss a dose, apply it as soon as possible and continue with your regular schedule. If it is almost time for your next dose, skip the missed dose and continue with your regular dosing schedule. Do not apply a double dose to make up for a missed one.If you are not sure what to do after missing a dose, contact your doctor or pharmacist for advice.
Store this medication at room temperature and keep it out of the reach of children.
Do not dispose of medications in wastewater or in household garbage. Ask your pharmacist how to dispose of medications that are no longer needed or have expired.
